All four myomiRs exhibited increased expression upon dihydrotestosterone treatment , hereby further suggesting that their genes are direct targets of the AR in muscle. Since expression of a large number of miRNAs in rat LA muscle is reduced by orchidectomy , muscle-specific miRNAs, also called myomiRs, are hypothesized to be mediators of myogenic androgen action. Conversely, decreased levels of putrescine, spermidine, and spermine have been observed in a rat model of muscle atrophy . The next section details this genomic action, including tethering of the AR by other transcription factors and androgenic regulation of polyamines and microRNAs in muscle.
Sarcopenic obesity means you have too little muscle and too much fat at the same time. So I care less about one universal perfect number and more about whether your score is low for your age and body size, or falling over time. In the European consensus on sarcopenia, low grip strength is treated as one of the first signs that something is wrong. In the PURE study published by Darryl Leong et al., lower grip strength predicted all-cause and cardiovascular mortality more strongly than systolic blood pressure.
Testosterone is an anabolic–androgenic steroid hormone produced mainly in Leydig cells of the testes in men and the ovary and the adrenal cortex in women. What is the relative importance of the many pathways that may cross talk with androgen action in skeletal muscle? A second approach to dissociate anabolic and androgenic activities of androgens is the development of tissue-selective AR-ligands, also called selective androgen receptor modulators (SARMs). However, the relative contribution as well as the exact mechanisms through which these non-genomic effects impact skeletal muscle growth and maintenance have to be further elucidated. Thus, myogenic androgen effects are partly mediated through non-genomic pathways (Fig. 2), including activation of a non-classical G-protein linked binding site on the plasma membrane of myoblasts resulting in Ca2+-dependent activation of several kinases. Using specific inhibitors, it is shown that the stimulation of L6 proliferation by testosterone involves the MAPK/ERK pathway, whereas PKA signaling plays a role in androgen-mediated stimulation of L6 differentiation .
Controls were healthy unrelated Russians without any competitive sport experience. Athletes were Russian national team members who had never tested positive for doping. They were classified according to their training frequency as mildly active (2 training sessions per week), moderately active (3–4 training sessions per week), highly active (5–7 training sessions per week) or extremely active (two training sessions per day).
